Abstract: 

One of the usages of SPERM cryopreservation is help to protect of endangered species. The life of SPERMatozoa is very short time in natural conditions and we can not conserve it for long time. While by semen freezing and use of suitable extenders and cryoprotectants we can conserve it in liquid nitrogen for several years and we can use stored SPERMs in essential times. In this research milt obtained from five Persian sturgeon (Acipenser persicus) were diluted using Tris-sacharose-egg yolk (23.4 mM Tris, 1.8mM sacharose, 20% egg yolk) as extender and methanol as cryoprotectant in four concentration (8, 10, 12, and 15%). The suspension was poured in to 0.5 ml straws and frozen in-196°C liquid nitrogen vapour in a three storage programe. Then straws were transferred to liquid nitrogen tank. The SPERM samples were thawed in a 40°C water bath for 25s and the SPERM MOTILITY rate and it's MOTILITY time were observed using microscope. There was not statistical significant difference between selected concentration (P>0.05) but the highest post-thaw MOTILITY rate and MOTILITY time (7.00±2.15% and 102.30±39.14s respectively) was found using 8% methanol after preservation in liquid nitrogen about 7 days while the lowest post-thaw MOTILITY rate and MOTILITY time (1.25±0.56% and 23.00±9.60s respectively) was found using 8% methanol after preservation in liquid nitrogen about 72 days. Results indicate that the increase in SPERM preservation in liquid nitrogen has a negative effect on SPERM quality and also increase in concentration of methanol from 8% to 15% does not change the percentage of motile cells and MOTILITY time on SPERM cryopreservation of Persian sturgeon. In This project methanol as a cryoprotectant had not successful efficiency on SPERM cryopresevation of Persian sturgeon.

Abstract: 

Magnetotherapy is a complementary therapeutic approach. It acts by applying the magnetic field and facilitates the improvement of the process. Infertility is one of the major clinical problems that affects about 10-15% of the world's population. Infertility in men accounts for about 40 to 50 percent. SPERM stimulation is one of the important parameters in male fertility. The aim of this study was to maintain the SPERM MOTILITY using a moderate intensity of static magnetic field. Materials and Methods: After initial studies, 90 male samples with normal SPERM specimens were collected. Each sample was divided into two parts, one specimen under the intensity of the magnetic field and one sample outside the magnetic range and both inside the incubator at1, 3 and 5 h intervals in the center of the static magnetic field. The total SPERM MOTILITY was determined by Casa software. Data was analyzed using SPSS software (version 16) and t-test. Conclusion: Static magnetic field can affect human SPERM MOTILITY. The results of this experiment showed that the magnetic field, after 5 hours, could keep the SPERM from moving outside the field.

Abstract: 

Background: The aim of this study was to evaluate the non-genomic action of thyroxin on SPERM kinetic and its probable use to improve SPERM recovery after applying an enrichment method like "swim-up" in comparison with the available one, pentoxifylline.Materials and Methods: This is an experimental study. A total of 50 patients were recruited, followed by infertility consultation. Conventional SPERM assays were performed according to World Health Organization criteria-2010 (WHO-2010). A Computer Aided Semen Analysis System was employed to assess kinetic parameters and concentrations. Number of the motile SPERM recovered after preparation technique was calculated.Results: Addition of T4 (0.002 mg/ml) to semen samples increased hyperMOTILITY at 20 minutes (control: 14.18±5.1% vs.17.66±8.88%, P<0.03, data expressed as mean±SD) and remained unchanged after 40 minutes. Significant differences were found in the motile SPERM recovered after swim-up (control: 8.93×106±9.52×06 vs.17.20×106±21.16×106, P<0.03), achieving all of the tested samples a desirable threshold value for artificial insemination outcome, while adding pentoxifylline increased the number of recovered SPERM after swim-up in 60% of the studied cases. No synergism between two treatments could be determined.Conclusion: We propose a new physiological tool to artificially improve insemination. The discussion opens windows to investigate unknown pathways involved in SPERM capacitation and gives innovative arguments to better understand infertility mechanisms.
